moulds_ Part 1: Colony count technique in products with
water activity greater than 0,95.
“This part of ISO 21527 specifies a horizontal method for the enumeration of viable yeasts and moulds in
products intended for human consumption or feeding animals that have a water activity greater than 0,95
[eggs, meat, dairy products (except milk powder), fruits, vegetables, fresh pastes, etc], by means of the
colony count technique at 25 ºC ±1 ºC.
This part of ISO 21527 does not allow the enumeration of mould spores. Neither the identification of fungal
flora nor the examinations of foods for mycotoxins lie within the scope of this part of ISO 21527. The
method specified in this part is not suitable for enumeration of heat-resistant fungi, such us, Byssochkamys
fulva or Bussochlamys nivea, in canned or bottled fruit and vegetables.“
Proceder à preparação da amostra de acordo com a ISO 6887-1.
Preparação da suspensão inicial da amostra numa proporção de 1:10;
Pesar 10/25g da amostra e juntar 90/225mL de Maximum Recovery
Broth (610077; 412400) ou Água Peptonada tamponada (611014;
414030; 414020) e se necessário proceder a diluições sucessivas.
Inocular pelo método de espalhamento 0,1 mL, da suspensão mãe e
respetivas diluições (em casos de amostras muito contaminadas,
selecionar para inoculação as 2 últimas diluições) e em placas com meio
de Dichloran Rose Bengal Chloramphenicol Agar-DRBC (610237; 81017;
10097).
Incubar a 25±1ºC/5 d
Identificar e contar até 150 colónias características ao fim de 2 e 5
dias
